Since the death of John F. Kennedy many theories have evolved regarding the individual or individuals who killed the president. Conspiracy theorist, as well as average citizens, believes Lee Harvey Oswald did not kill the president, or that he was not the only person involved in the assignation. My research would attempt to design a chip that could process data faster, and with more accuracy, than is presently possible. This chip could then handle more complex software, process data from CDs, and handle Internet applications at a faster rate, allowing for equalization with the increase in speed of CD ROM drives and modems. The procedure for this research would entail first learning as much as possible about the design of present chips, the specific materials that I might want to include as conductors, and which metals are easiest to manipulate. Next I would lay out the chip and attempt to construct it with existing technology. Finally, I would need to test it in an effort to make modifications and improvements in the original design. The computer age presents wonderful opportunities, and any research that promotes the facile use of PC or network systems will help advance this cause. For this reason, I would dedicate my year of research to improvements in the design of central processing units.

Most of the Sahara Desert is undeveloped and features a varied topography. Most of its landscape has been shaped over time by wind and includes sand dunes, sand seas called ergs, barren stone plateaus, gravel plains, dry valleys, and salt flats. Around 25% of the desert is sand dunes, some of which reach over 500 ft (152 m) in height. There are also several mountain ranges within the Sahara and many are volcanic. The highest peak found in these mountains is Emi Koussi, a shield volcano that rises to 11,204 ft (3,415 m). It is a part of the Tibesti Range in northern Chad. The lowest point in the Sahara Desert is in Egypts Qattara Depression at -436 ft (-133 m) below sea level. Most of the water found in the Sahara today is in the form of seasonal or intermittent streams. The only permanent river in the desert is the Nile River that flows from Central Africa to the Mediterranean Sea. Other water in the Sahara is found in underground aquifersà and in areas where this water reaches the surface, there are oases and sometimes small towns or settlements like the Bahariya Oasis in Egypt and Ghardaà ¯a in Algeria. Since the amount of water and topography varies based on location, the Sahara Desert is divided into different geographic zones. The center of the desert is considered hyper-arid and has little to no vegetation, while the northern and southern portions have sparse grasslands, desert shrub and sometimes trees in areas with more moisture. For example, during the last glaciation, it was bigger than it is today because precipitation in the area was low. But from 8000 BCE to 6000 BCE, precipitation in the desert increased because of the development of low pressure over ice sheets to its north. Once these ice sheets melted, however, the low pressure shifted and the northern Sahara dried out but the south continued to receive moisture due to the presence of a monsoon. Around 3400 BCE, the monsoon moved south to where it is today and the desert again dried out to the state it is in today. In addition, the presence of the Intertropical Convergence Zone, ITCZ, in the southern Sahara Desert prevents moisture from reaching the area, while storms north of the desert stop before reaching it as well. As a result, the annual rainfall in the Sahara is below 2.5 cm (25 mm) per year. In addition to being extremely dry, the Sahara is also one of the hottest regions in the world. The average annual temperature for the desert is 86à °F (30à °C) but during the hottest months temperatures can exceed 122à °F (50à °C), with the highest temperature ever recorded at 136à °F (58à °C) in Aziziyah, Libya. There are plenty of reasons why it is better not revealing information. Withholding information that may put the population in danger would be better than revealing information which would bring chaos. On the flipside, transparency and honesty between the government and the public are very important. Transparency is essential in order for a democracy to function and honesty is important in cases when there is need for strong relations to be created. And although I support the idea of freedom of information, the general public will never be educated enough to maintain consensus and national problems. In these cases withholding information helps to ensure stability in terms of national security, preserving the peace and well-being. The main reason authority keeps information is the fear from criticism and disapproval. A rational society would never be satisfied with vague information and would want to take part in the decision-making process. Government has a lot of information which the people may have an interest in but still some of this information is dangerous. Things like war strategies and secret missions need to be withheld. Being kept in the dark on some things as long as its truly for the good of the nation and not for someoneââ¬â¢s benefit is acceptable. For example if government find the cure for cancer but decide that it would ruin pharmaceutical companies and chose not to reveal this information it isnââ¬â¢t right. On the other hand, there is a lot of information which public doesnââ¬â¢t deserve access to. Like information that endangers the government. Another reason for withholding information could be to cover mistakes and not causing discontent. The government always makes big promises and eventually it will have to fulfill some of them. If government promise to reduce the crime rate in 30% within a year and donââ¬â¢t follow the plan it will lose itââ¬â¢s credibility. I think that the idea some information to be withheld is valid. However, information that would improve the living standard of people or information which is of great importance for the future of humanity, in my opinion shouldnââ¬â¢t be kept. There is no clear moral answer here. When there is too much that is at stake, we canââ¬â¢t say that information should always be free. Should it benefit the public to know how their government works, or it should benefit the government to have the full control over our lives. Im not so sure.

Fiber optics was developed by Bell labs and Corning in the late 1960s. It does not experience signal degradation over distance as would coaxial cable. School districts are aware of the need for the upgrades to fiber optic cabling, but costs frequently preclude the upgrade. Federal Communication Commission (FCC) rulings in October of 2004, that relieved incumbent local exchange carriers from having to share fiber networks that reach within 500 feet of homes, have led to plans by BellSouth to boost fiber deployments. Concern among competitors is that their ability to compete for business voice service will be hurt. (Quesada, 2004). But while the unbundling protection for fiber-to-curb is a blow to competitive local exchange carriers, BellSouth plans to increase deployments of fiber-to-the-curb by 40 percent in 2005, a move that will help decrease the cost for local school districts. Since any installation of new wiring is labor-intensive and costly, it is little wonder that school districts lag behind the corporate world in obtaining this superior technology. School districts are rarely provided with enough state and local funds to expand current technologies to encompass such upgrades. There are numerous programs and opportunities in place, however, that will allow even less affluent school districts to keep pace. One such program is called the Universal Service Fund for Schools and Libraries, or more simply, ââ¬Å"E-rateâ⬠. E-rate was created by the Telecommunications Act of 1996, and is the discounted rate that schools and libraries pay for access to affordable telecommunications services. It was passed with the help of such politicians as Bill Clinton, Al Gore, and Newt Gingrich. The Act gave the Federal Communications Commission the power to expand universal access so that schools and libraries would be Internet wired. (Carolan & Keating, 1999). Since then, the trend to sign up for E-rate has swept the country. In 1999, Merrick and North Babylon students in Long Island, New York, discovered internet access when their schools opened, and Massapequa voters approved a multimillion-dollar bond issue that included Internet connection costs. Professional Development Plan Essay
My professional plan is to grow as a nurse by gaining knowledge through school and my daily experiences, so in five years I can care for patients and families as a nurse practitioner. I will make short term and long term goals to help guide me down my path as I transition into the role of professional nurse; providing me with new opportunities for my future. Beliefs and values influences the decisions people make in their personal and professional life. My beliefs and values were incorporated to make my philosophy of nursing, that is to be an advocate for my patientsââ¬â¢ and their family during their most vulnerable time, continuing my education to provide the best quality care available and taking a holistic approach by looking at the person as a whole; caring for their body, mind, and spirit (Chitty, 2007 p. 312). I believe nursing is an evidence based practice, the care provided to patients and family have been proven through research to be the safest most effective way to implement care. My values which mold my beliefs have been acquired throughout my life from the people around me and my experiences. My values and beliefs will continue to change as I get older, have more experiences in my profession and further my education in nursing. One of my short term goals is to finish my baccalaureate degree in nursing. In terms of advancing professionalism, research was recently published, (Morris & Faulk, 2007), in which nurses were found to have increased professionalism and growth in roles and values after attending an ADN to BSN program. That is what I am striving to achieve while going through the BSN program. I plan on taking each class one at a time, trying to do my best, and gain the knowledge needed to grow in my field. By August 2009, I should have my BSN which will provide me with new opportunities such as management and educator positions; this degree will also provide me the opportunity to further my education to a masterââ¬â¢s level. My second short term goal will begin this summer, that is to initiate the evidence based practice, EBP, project on my progressive care unit, PCU, at Banner Good Samaritan Medical Center, BGSMC. Several of my coworkers and I are currently gathering information on research that suggests that providing patients and their families with written and oral information on the side effects of their medications will improve patient satisfactory scores in a acute care setting. We meet together regularly, learning as a team how to initiate EBP into the workplace. Then possibly in the future we will publish our findings. By fall of 2009 I plan on starting the Masters of Science in Nursing/Family Nurse Practitioner program at the University of Phoenix so I will be able to sit for the national boards certifying exam by the end of 2012. Once I complete the requirements I must submit the documentation to the Arizona State Board of Nursing. I then plan on working for an interventional cardiologist, which will give me the opportunity to work in both the hospital and office setting. Income is not the sole reason for my desire to advance in my profession but it is important to note that the U.S. Census Bureau 2000 data show that income levels rise as educational levels rise (Katz, Carter, Bishop and Kravitz, 2004). Income will enable me to continue my education and to obtain other goals in my future such as going to law school and traveling around the world. When discussing a professional development plan people have to keep in mind a persons support system, resources available to them, their work ethics and what is needed to succeed. While one may be able to fulfill his or her goals without one or more of these things, adversities can deter many from following plans they set forth. Support and understanding from my family and friends would be a significant tool in helping me achieve my goals. It may also be useful to have mentors help to guide me along my chosen path to success. Financial assistance will also be needed to continue on with my learning process. Self-discipline, further education and experience are also required to be successful in my future role as an FNP. Self-discipline and time management skills are crucial to have in our fast pace speed of livesââ¬â¢ today. It becomes increasingly important in the studentsââ¬â¢ life to use time wisely. Students may have to overcome several hurdles and barriers along the way. It is important to be able to identify these issues; it is the first step to overcoming them. Barriers may be financial, time restraints or lack of support from family and friends; having the knowledge how to overcome these barriers and having backup plans is what makes the difference between meeting your goal versus abandoning the goal. If faced with any of these barriers I will try to have a positive outlook and attitude, I will try to look on the bright side. Michaud (2002) observed that oneââ¬â¢s ability to bounce back from disaster and loss depends on the personââ¬â¢s ability to use resiliency skills to conjure up positive emotions when he or she is challenged severely. I will monitor my stress level and use relaxation techniques specific to me and my needs if necessary in order to avoid abandoning the goals that I have set. In conclusion, putting together a professional plan is an important step to obtaining desired outcomes. According to Alexander Graham Bell (2001) ââ¬Å"before anything else preparation is the key to successâ⬠. Values and beliefs will change over time, which will influence your original plan to formulate new goals, when this occurs a person must revaluate the resources available and challenge themselves to meet new goals. It has been taught in American schools that the war fought in order to stop slavery and that the American South did the right thing by seceding from the Union because of its support in slavery. Subsequently, the Civil war was a result of the Founding Fathersââ¬â¢ failure to address the institution of slavery in a republic that proclaimed in its Declaration of Independence that all men are created equal. Nonetheless, the necessity of the war has been questioned because the four-war to abolish slavery ravaged the country and destroyed nearly half of the American people. Among the countries that practiced slavery, only America and Haiti freed their slaves by war. The rest were freed peacefully. Hence, it can be concluded that the South could have gained its independence without the war. The rising alone of keeping and posting guards on the constantly escaping slaves would have resulted in the gradual decline for the demand of slaves. The absence of a fugitive slave laws in other states that would force the return of the slaves to their owners has added to the decreasing value of the slaves. As such the institution would have been unsustainable regardless of the moral and humanitarian perspectives. On the other hand, the American North would still have achieved its goals without resorting to war. Apparently, the North slaughtered a large number of American young men in the battles against the South. Nonetheless, it is evident that the South posed no military threat to the North. Its aim of preserving the Union was considered as not a compelling reason to continue the war. The North wanted to keep the Union intact solely on the basis of economy and fiscal matter. Since the principal source of tax revenue for the federal government before the Civil war was a tariff on imports, they needed to keep these tariffs in which the federal government accounted its federal budget. The North did not need to take the bait by Lincoln who dispatched southerners to forestall the supplies for Fort Sumter. Hence, its attack on the South could have been prevented had not depended on the Union being intact and the economic advantages it provided them.

Bilingual education was first initiated in 1968. It was a new means to educate the children who spoke a minority language. thirty-one years later the same problems exist for those children who speak a language other then English. The experiment of Bilingual education has been a failure and now itââ¬â¢s time to move on. The first English only initiatives were brought forth in 1981 by newly elected president Reagan. Since then the conflict over Bilingual education has drove on. Currently twenty-three states have ââ¬Å"English Onlyâ⬠laws with 4 states having laws that are pending. The issue of bilingual education and the conflict that surrounds it is primarily focused between whites and Hispanics, although since the mid 70ââ¬â¢s itââ¬â¢s also been involved with the Chinese and Mung cultures. Bilingual education has failed however, currently the movement is towards and English only educational system. The simple fact of the matter is that people who speak a language in th is country will never get ahead. Weââ¬â¢ve tested the ignorant notion that you could get by without speaking English; or speaking English very minimally, but thatââ¬â¢s promoting and even more ignorant idea which is that you donââ¬â¢t need speak English fluently to succeed in America. In 1968 we didnââ¬â¢t know that Bilingual Education wouldnââ¬â¢t work, however, itââ¬â¢s time to move on and do the right thing and switch to English Only. à à à à à Lyndon Johnson was president, we had the futile dream of world peace, at the same time we were ââ¬Å"Bombing the North Vietnamese into the stone-ageâ⬠. In the final year of LBJââ¬â¢s presidency sever new education initives were taken as part of the last step in his ââ¬Å"Great Societyâ⬠programs. One of these new initives was Bilingual education. At the time it was supported by the Liberal whites in this country, for the most part the conservative Hispanic sect was opposed to such measures. The idea at the time was, kids would learn in their native language and simply pick up English gradually. Those ideas were radicalized in the 1970ââ¬â¢s however, the premise moved away from gradual learning of English; to English wasnââ¬â¢t really necessary. In 1981 Bilingual education came under assault from newly elected president Reagan proposed moving to the English Only system. The idea wasnââ¬â¢t viewed as Anti-Hispanic at the time, or as some kind of racist proposal. Reagan was adored by a majority of th... ...and since 1908 printed exclusively in English by the United States Government. Teddy Roosevelt said in 1908 that ââ¬Å"To print the American Constitution in any other language but English, would be like spitting in the face of our forefathersâ⬠. The Untied States Supreme Court has also ruled numerous times that English only laws do not violate the 1st amendment of our Constitution. à à à à à I personally believe that in our nation there needs to be a main language, not 3 or 4 or 1200. George Washington wasnââ¬â¢t giving orders at Valley Forge in Russian he was speaking English. Abe Lincoln didnââ¬â¢t give the Gettysburgh Address in Portuguese. Itââ¬â¢s not to much to ask for every American citizen to be able to speak English fluently. No one is forced to live here, and if you refuse to adapt you probably shouldnââ¬â¢t be living here. If you want to speak German by all means go ahead and do so, but do it back in Germany. While youââ¬â¢re in America however, we speak English, and if you donââ¬â¢t know it youââ¬â¢re in trouble. Favorite Family Get-Together
Getting together with family is something that most of us look forward to. Throughout the years our family has grown with all sorts of personalities, Making every get-together fun, interesting, and memorable. Even though I see my family quite often, our get-togethers are always quite entertaining. While we celebrate birthdays, holidays, special occasion, and new or old traditions, By far my three favorite are my mom's birthday, pumpkin carving contest, and New Year's.My first favorite family get together is my mom's birthday. No matter what day the week her birthday is my siblings and I always managed to gather our families enjoying my mom for dinner. It's indescribable how happy that makes her. We bring her favorite dish and pastry. At the dinner table we take turns to express our gratitude and I love for my mom. Throughout the night we gathered up and see her open the gift and read the card her grandkids made for her. Every year is a blessing to be able to enjoy that moment.My seco nd favorite family get together is our pumpkin carving contest. We came up with this tradition couple of years ago. Teams are formed by family and their kids, we help them carve their pumpkin into any character or picture of their choice. To make it fun we give out prices for the winning pumpkin. The children get so involved and become competitive that the competition gets quite interesting. The losers end up cleaning the mess left over from the carving process it became more popular with the kids as they grow older.The competition became more serious. But at the end of the contest everyone goes home with a great carved pumpkins. My third favorite family get together with the New Year's day. Our family celebrates not only for the upcoming new year But also for the year that ends. The children play throughout the night and the adults just relax, Conversation; Making it seem like we rarely see each other throughout the year. My family only consists of my immediate family.Making it mor e meaningful, private, and warming. The countdown to the new year is by far the best. We get in spirit, putting on hats, glasses, beads of the upcoming new year numbers. As a family we end the year and we welcome the new year with popping streamers, and loud horns, The most importantly as a family. Throughout the years our family get-togethers have become memorable. Celebrating birthdays traditional events and holidays makes our little families become one.

Title: DNA analysis Aim: a) Isolate and Purify Bacterial Chromosomal DNA from a strain of E. coli b) Visualization of restriction fragments by Agarose Gel electrophoresis Objectives: * to isolate and purify bacterial chromosomal DNA from a strain of E. coli * to analyze and identify DNA by use of a spectro-photometer * to use restriction enzymes to cleave DNA into fragments * to visualize the restriction fragments by gel electrophoresis * to compare the different DNA fragments generated by use of molecular markersAbstract This work describes a lysis method for the isolation and purification of bacterial genomic DNA and visualization of the restriction fragments by agarose gel electrophoresis. It was noted that for one to isolate and purify bacterial chromosomal DNA several steps are taken into consideration. DNA was found to absorb at 260nm wavelength in a UV spectrophotometer. Restriction enzymes were added to cleave DNA which would produce various DNA fragments. DNA can be separate d into different sized fragments by gel electrophoresis.The bacterial DNA was successfully isolated and purified however it could not be observed after running the gel. DNA analysis is a standard practice for defining paternity or maternity, predisposition to disease, embryonic health and criminal guilty. But in our context, DNA analysis is mainly used for predisposition of diseases in bacteria. Bacteria are pathogenic microorganisms that cause infectious diseases including cholera, syphilis, anthrax and leprosy. The most common fatal bacterial diseases are respiratory infections such as tuberculosis (Barnum S.R; 1998). Nucleic acids encode information relating to cell structure and function. Cells have the ability to make copies of their DNA and pass this information to daughter cells. Nucleic acids are polymers of nucleotides. Nucleotides are composed of ribose (a 5` carbon) sugar and either a purine and pyrimidine base at 1` position. The purine bases are adenine (A) and guanine (G) and the pyrimidine bases are cytosine (C), thymine (T) and Uracil (U). Uracil is only found in RNA and thymine is only found in DNA (Wiser M. F; 2002).Isolation of nucleic acid ââ¬â three major types of techniques are employed in the isolation of nucleic acids differential solubility, absorption methods or density gradient centrifugation. The choice of method will depend on the type of DNA being isolated and the application. A major goal of nucleic acid isolation is the removal of proteins. The separation of nucleic acids from proteins is generally accomplished due to their different chemical properties. In particular, the highly charged phosphate backbone makes the nucleic acids rather hydrophilic as compared to proteins which are more hydrophobic (Allison L.A; 2012). Spectrophotometry is a versatile analytical tool. The underlying principle of spectrophotometry is to shine light on a sample and to analyze how the sample affects the light. DNA absorbs light at a wavelength of approximately 260nm (Stryer; 2006). Centrifugation is a process that involves the use of the centrifugal force for the separation of mixtures. Separation is based size, shape and density. It utilizes density difference between the particles/macromolecules and the medium in which these are dispersed (Gupta P. K; 2006).Dispersed systems are subjected to artificially induced gravitational fields. A buffer is an aqueous solution consisting of a mixture of weak acid and its conjugate base or weak base and its conjugate acid. Its pH changes very little when a small amount of strong acid or base is added to it and thus it is used to prevent any change in the pH of a solution (Cowan M. K; 2009). Electrophoresis is a diverse technique of separation used to separate and sometimes purify macromolecules especially proteins and nucleic acids that differ in size, charge or conformation by an electric current (Stryer L. 2006). Gel electrophoresis refers to using a gel as an ant convective mediu m and or sieving medium during electrophoresis. Gel electrophoresis is most commonly used for separation of biological macromolecules such as deoxyribonucleic acid (DNA), ribonucleic acid (RNA), or protein; however, gel electrophoresis can be used for separation of nanoparticles. Materials Used * Luria Broth medium * SET Buffer * TEN Buffer * Choloroform/isoamyl alcohol. 24:1 mixture * Phenol/ chloroform 1:1 (Buffer saturated phenol) * Ethanol (95%) stored at -20? * Na Acetate * NaCl: 5M sterilized by autoclaving Sodium dodecylsulphate (SDS) : 26% (w/v) * Bacteria cells * Plastic test tubes * Glass rods * Wide bore pipette * Ice bath * Centrifuge * Ethidium bromide * Agarose * TBE buffer Methodology Each group carried out the following procedures: Used two 50ml sterile plastic tubes, harvested cells by centrifugation for 10 min 4ââ¬â¢C. Combined pellets to give approximately 1g wet weight of cells. Washed the pellet, re-suspended it in 20ml Ten buffer by gentle vortexing. Harvest ed the cells again as described above. Re suspended the cells in 10ml of Set buffer and let them sit on ice for 5min.Added 1000à µL of lysozyme and incubated at 37? for 30 min. Divided the cell suspension into two in separate sterile 50ml tubes. Added 5 ml Ten Buffer and 500à µl of SDS. Gently mixed the tubes by inverting them until lysis occurred. To each tube added 1ml 5M NaCl and an equal volume of buffer saturated phenol. The tubes were inverted till the mixture was emulsified. Separated the phases by centrifugation for 10min at 40C. Recovered the upper aqueous phase using a wide bore pipette. When retaining the aqueous phase the pellicle at the interface was avoided. Repeated the extraction until the interface was clear.Added an equal volume of chloroform and extract residual protein as described above. Transferred the upper aqueous phases from both tubes to a 100ml beaker. Set them on ice and added 1/10th volume 3M Na acetate. Precipitate the DNA by addition of 2 volumes of ice cold 95% ethanol. Mixed thoroughly and allow it to stand for about 5min on ice for the DNA to precipitate. Spooled the DNA out of solution on a glass rod, dipped it into a tube of 95% ethanol and re-suspended in 10ml Ten Buffer. Left to dissolve overnight at 4ââ¬â¢C B) Gel electrophoresis The gel was prepared by melting 1. 6g of agarose plus 200ml of 0. x TBE buffer. Swirled the mixture and allowed it to cool to 55?. Added 10? l ethiduim dye Loaded the gel in the following order; 1. Undigested pBSK 2. pBSK + digested with Eco R1 and Xba 1 3. Undigested DNA from a blue colony 4. DNA from a blue colony digested with Eco R1 and Xba 1 5. Undigested DNA from a white colony 6. DNA from a white colony digested with Eco R1 and Xba1 7. Lambda Hind III molecular weight markers After loading the gel it was run at 100 volts for 2 hours. Results We managed to precipitate DNA out of the Bacterial cells. DNA was seen a small white like fragments.However we could not spool the DNA out of sol ution using glass rods due to fact that DNA is a fragile compound hence when we twisted / spooled for DNA we destroyed the DNA strands cutting them into smaller fragments. The following day, analysis of the DNA sample in a spectrophotometer was carried out. It was found that DNA absorbed a specific wavelength of 260nm. This proved the presence of DNA in the sample. Our sample was digested by restriction enzymes and labeled the DNA fragments with an identification dye and ran them on the Gel electrophoresis together with molecular weight markers.After running the gel no observeable bands of different band fragments were observed. Only the molecular weight markers bands were observed. Discussion The TEN and SET buffer were used to lyse the cells. They are good buffering agent, which solubilizes the DNA, while protecting it from degradation. Eluting and storing the DNA in TBE Buffer is helpful if the EDTA does not affect the downstream applications. EDTA chelates or binds to Mg2+ ions present in purified DNA and can help inhibit possible contaminating nuclease activity (Cowan M. K; 2009).Balancing of test tubes before centrifugation in order for the centrifugation process to be effective to create centrifugal field that results in maximum separation of cell components. According to Wiser M. F 2002, DNA is very insoluble in ethanol and isopropanol, but both alcohols are very water soluble. Thus, it will dissolve in water to form a solution and cause the DNA in the solution to aggregate and precipitate out. Isopropanol is often better to use because it has greater potency in precipitating the DNA and thus lower concentration is required. This is advantageous because it will take less time for the isopropyl alcohol to evaporate.Salts such as sodium chloride and ammonium acetate remove histone and non-histone chromosomal proteins bound to the DNA. As soon as 95% ethanol was added after sodium acetate for DNA precipitation, the whole solution turned cloudy with a lot of white precipitate, precipitating down. According to Allison L. A, 2012; sodium acetate which is negatively charged and low pH was used which contributes to charging positively the DNA. A combination of this plus high salt molarity enhances formation of aggregates of DNA and facilitates the pelleting procedure. Chloroform isoamyl-alcohol is a type of detergent.It binds to protein and lipids of cell membrane and dissolves them. By this it disrupted the bonds that hold the cell membrane together and cause it to breakdown. It then forms complexes with these lipids and proteins, causing them to precipitate out of solution (Besty T and Keogh J; 2005). This reduced chance of contaminated DNA being obtained hence making it possible for us to be able to precipitate DNA only. Alcohol (95%ethanol) is used to precipitate DNA. SDS which stands for ââ¬Ësodium dodecyl sulfate' is a strong anionic detergent that can solubilize the proteins and lipids that form the membranes.This will helped t he cell membranes and nuclear envelopes to break down and expose the chromosomes that contain the DNA. In addition to removing the membrane barriers, SDS helped release the DNA from histones and other DNA binding proteins by denaturing them (Barnum S. R; 1998). Ethidium bromide is an intercalating agent commonly used as a fluorescent tag (nucleic acid stain) in molecular biology laboratories for techniques such as agarose gel electrophoresis. When exposed to ultraviolet light, it will fluoresce with an orange colour, intensifying almost 20-fold after binding to DNA (Wiser M.F; 2012). Molecular weight size is a set of standards that are used to identify the approximate size of a molecule run on a gel. These markers were composed of nucleic acids of different sizes. A few reasons you may not see bands on the gel after electrophoresis: When preparing the gel for electrophoresis TBE buffer was used. This was done so that the temperature can be maintained and lubricate the electrolyte. L oading dye was added this helped weigh down the DNA so that it can sink into the bottom wells and not float in the buffer solution. According to Gupta P.K, 2006; loading dye moves quickly than the actual DNA parts so it is an indicator to when to turn off the power on the electrophoresis chamber. The dye also makes the DNA visible to the naked eye, giving it a purplish color and making it easier to work with. After Gel electrophoresis no bands of DNA were observed. This according Allison L. A (2012) might have been as a result of any of the following * DNA concentration might have been too low. * DNA sample is contaminated with RNA and Protein * DNA bands are too small and have run out of the gel The buffer system in which the gel is suspended is not doing its job correctly. The buffer might have to be made fresh. * The electrophoresis apparatus is not in the correct orientation (electrodes not connected to the right poles). The major drawback in the experiment was that our fellow c olleagues were not able to isolate and purify their DNA. Also when working with DNA temperature regulations were not sometimes adhered to, it was sometimes left on the surface tables for long periods esp. when the samples were being analyzed in the spectrophotometer.Recommendations With proper teamwork and co-ordination among my fellow classmates much larger quantities of DNA could have been isolated and purified. The DNA should not be kept at room conditions for a long time. Conclusion The experiment was partly a success managed to isolate and purify DNA, analyzed it using a spectrophotometer.
